"It is creating quite a stir," said cathedral chief executive Roy Bentham.
"A lot of preparations have gone into it. It is a huge job in terms of organisation."
Cathedral bosses will also ask for a Renaissance painting by Titian pupil Paris Bordone to be valued on the show.
Bordone's painting shows the Holy Family with St Catherine and was donated to Lincoln Cathedral by Dorothy Hood in 1963 after being bought by her father in 1885.
Mr Bentham said: "Obviously it is an important work of art and we are intrigued to know more about it and are interested in its value."
Bordone worked in Venice in the 1500s and as well as some surviving canvas works, he also painted murals, all of which are now lost.
The BBC's specialists welcome anybody with a family heirloom or a vase which has done nothing but gather dust for decades.
The filming of this first episode in the new series of Antiques Roadshow will last all day, starting from 9.30am.
Show bosses confirmed that as long as Lincolnshire fans join the horde of would-be antique owners by 4.30pm they stand a good chance of having their prized possessions valued.
