The company, which has 133 stores in the UK, says it sells a range of 20,000 products for the home.
The new store, which replaced the firm's old property at a separate site in Outer Circle Road, also has a coffee shop.
Chief executive Nick Wharton said: "We are always excited about the opening of a new store.
"We are confident that Lincoln will continue to play a big part in the continued success of the company."
Bill and Jean Adderley founded Dunelm Mill in 1979 as a market stall business in Leicester selling ready-made curtains.
The first store was opened in 1984 and since then the business has developed into a successful nationwide chain.

“I only went to Dunelm Mill once. The music was so loud I left after 5 minutes. Never going back. I like to shop in peace and quiet.”

“Do the 50 new jobs include the people who were employed at the old shop? If so how many TRUE new jobs have been created. I expect most of them are part time anyway!”

“The only problem I can think of is trying to find somewhere to park in the Carlton Centre car park now. Will the car park be big enough to cope with a store of this size along side the other popular businesses up there?”
